Application of mass spectrometry to the study of differential protein expression (proteomics), enzyme mechanisms, macromolecular structure, and carbohydrates in disease

Major research interests in Dr. Gibson¹s laboratory encompass the growing intersection of modern mass spectrometry and structural biology, with current projects including studies on the molecular events underlying bacterial pathogenesis, mechanisms of oxidative stress in neurodegenerative diseases, and tertiary protein structure determination. These efforts all rely on the structure determination of important bio-molecules and the continued development and application of advanced mass spectrometry and other bio-analytical techniques.

Current active projects in Dr. Gibson group involve five major research areas;

1) structural, biological and immunochemical studies of the role of surface carbohydrates (lipooligosaccharides, or LOS) in pathogenic Neisseria and Haemophilus spp.,

2) the development of a carbohydrate-based vaccine and other therapeutic strategies,

3) the mechanism of iron-acquisition and iron-regulation in Mycobacterium tuberculosis and the identification of iron-regulated proteins,

4) the role of oxidative stress mediated through mitochondrial dysfunction and its contribution to altered protein expression (Œproteomics¹) and oxidative modification of DNA and proteins in Alzheimer¹s disease, and

5) the development of a mass spectrometry-based method using chemical crosslinking and computation for the determination of the tertiary structures of proteins.
